CSL tune, S5 pretty much all the time.

Auto 3, so others can drive the car (kids, mom), Manual 5 or 6 for me, exclusively, usually 6 just because when I want to drive the damned thing, I don't need the nanny controls.

After CSL tune, I use S4 daily and S5 when I am driving spirited or at the track. I used S6 at my last track event and found that it was almost too aggressive and would sometimes upset the car a bit. S5 is almost just as fast but does a better job at keeping things smooth. However, if I were drag racing then I would use S6.
